Obesity is a nutritional disorder being a global problem that affects more than 650 million people. Obesity is associated with the development of endoplasmic reticulum stress (ERE) and the activation of responses to stress in metabolically active tissues. ERE in the adipocyte is produced as a consequence of an excess of fatty acids and glucose, which affects the functional homeostasis of the ER. In recent years, the use of polyphenols to regulate ERE and reverse metabolic disorders associated with obesity is increasing. Studies by this group of investigation have shown that the LC-HS extract mixture improves the lipid profile, glucose levels and tolerance, reduces the expression of pro-inflammatory cytokines, etc. However, it is unknown the if it could also modulate metabolic stress through ERE inhibition. Therefore, the objective of the present work was to determine the effect of the LC-HS and their metabolites on ERE and apoptosis in hypertrophic 3T3-L1 adipocytes through the determination of the expression and / or activation of various ERE markers. The expression of pIRE1-α / IRE1-α, ATF6, PERK, CHOP and PARP was evaluated because they are the main activation pathways of UPR during ERE, in the case of CHOP and PARP, was because they are markers of apoptosis during ERE. Finally, it was verified if these modulation in the expression of markers translates into changes in ER mass in a model of hypertrophic adipocytes induced by high glucose concentration. The extract and some metabolites exerted a negative regulation in the expression of these markers and the mass of the ER was significantly reduced. Therefore, these results can be used for the potential use of these compounds as modulators of the proteins studied and their implication in the metabolic pathways as well as the effects of these compounds in the amelioration of the obese condition.
