Transient potential receptors (TRPs) are a group of eight ion channel subfamilies based on
sequence homology and cation permeability, which play critical roles in the responses of
external stimuli. This study is focused on TRPM8, a well-known thermoregulator channel
involved in sensory transduction of different stimuli. Several studies have found that the
TRPM8 dysregulation is associated with diverse pathologies development. Although all the
research advances, TRPM8 specific modulators still are a clinical need. Therefore, the objective
of this study was to characterize eight compounds derived from tryptamines, chemically
synthesized, as modulators of TRPM8, through screening, potency, cytotoxicity, and specificity
tests by in vitro techniques. Compounds with the best pharmacological properties were C5
(IC50= 0.4 ± 1.5 μM) and C8 (IC50 = 0.4 ± 1.1 μM). All tryptamine derivatives tested induced
specific antagonism in TRPM8, which highlight the relevance of this configuration in future
modulators development. Finally, the complete characterization of best antagonists by
electrophysiological characterization it´s the key for further development into preclinical and
clinical studies.
