The human are able to detect and prevent tissue damage through nociception, which is the process by which the primary afferent (sensory) nerve fibers of the somatosensory system detect harmful stimuli. These afferent nerve fibers are known as nociceptors or pain receptors. The excitability of these nociceptive neurons is regulated by different ion channels. Both these nerve endings in charge of capturing and processing the pain signal, as well as the various ion channels that regulate its activity, have become therapeutic targets for the purpose of creating new drugs against pain. In this study the cell line MED17.11 has been differentiated to nociceptors. The expression of mRNA of several neuronal markers has been investigated by RT-PCR techniques, although expression of adult nociceptor markers has not been observed; and the functionality of the TRPV1 channels was examined by calcium image, obtaining few response to capsaicin and ATP but without response to KCl pulses, indicating that differentiated neurons lack in terms of action potential neuronal functionality. To achieve expression of adult nociceptor markers as well as stable neuronal functionality, it will be necessary to customize the differentiation protocols for achieving proper nociceptor differentiation.
