The ribosome is a universal and essential machinery whose biogenesis consumes the majority of the energy resources of any cell and, therefore, it is strictly regulated. We are studying several Arabidopsis genes encoding putative ribosome biogenesis factors (RBFs): PLANT RNA HELICASE75 (PRH75), U3 SMALL NUCLEOLAR RNA-ASSOCIATED PROTEIN 18 (UTP18), RIBOSOMAL RNA PROCESSING 36 (RRP36) and POLYMERASE V (POL5). No functional studies of these genes have been published in Arabidopsis, except for PRH75, which participates in 18S rRNA maturation. To determine the subcellular localization of these putative RBFs, we generated transgenes containing translational fusions of these genes and the GFP gene, driven by their endogenous promoters. We transferred the transgenes to POL5/pol5-1, UTP18/utp18-1 and prh75/prh75 and their respective wild-types. The PRH75pro:PRH75:GFP and UTP18pro:UTP18:GFP transgenes partially rescued the atrh7-2 and utp18-1 mutant phenotype, respectively. However, the POL5pro:POL5:GFP transgene completely rescued the embryonic lethal phenotype of the pol5-1 mutant. Furthermore, we have determined that all four proteins are predominantly nucleolar, which was expected.
