Abstract:
Existen aproximadamente 10 millones de personas alrededor de todo el mundo que viven con diabetes tipo 1. Con el fin de mejorar las terapias clásicas, se han abierto nuevas líneas de investigación que buscan alternativas para la prevención del desarrollo de diabetes tipo 1. Una de las líneas de estudio más importantes es la vía de señalización de IFNα. En este trabajo se pone a prueba zasocitinib, un inhibidor específico de TYK2 en fase clínica II para el tratamiento de psoriasis, como una posible terapia para la prevención de los daños inducidos por IFNα en célula β pancreática. El presente trabajo demuestra que zasocitinib protege a las células β de la apoptosis inducida por IFNα e IL-1β a través de la inhibición de la vía señalización de IFNα, mediada por la fosforilación de STAT1, entre otros factores. Además, su efecto protector se establece por su capacidad de prevenir la sobreexpresión de HLA de clase I y genes proinflamatorios. Sin embargo, zasocitinib no es capaz de prevenir la sobreexpresión de marcadores de estrés de retículo. Los resultados obtenidos con zasocitinib lo postulan como un gran candidato para evitar los efectos adversos inducidos por IFNα en la fase temprana de diabetes tipo 1.
There are approximately 10 million people around the world that live with type 1 diabetes. With the objective of improving classic therapies, new research topics have opened new avenues to seek alternatives for the prevention of type 1 diabetes development. One of the most important investigation points is the signaling pathway of IFNα. In this study, we have tested zasocitinib, a TYK2 specific inhibitor in clinic phase II for psoriasis treatment, as a possible therapy for the prevention of the damages produced by IFNα in pancreatic β cell. The present investigation demonstrates that zasocitinib protects β cell from the apoptosis induced by IFNα and IL-1β throughout the inhibition of the signaling of IFN, mediated by the STAT1 phosphorylation, besides other factors. Moreover, its protectory effect is stablish by its capacity to prevent the overexpression of class I HLA and proinflammatory genes. Nevertheless, zasocitinib cannot prevent the overexpression of ER stress markers. These findings suggest that zasocitinib may be a great candidate to avoid the adverse effects induced by IFNα during the early stages of type 1 diabetes.
