dc.description.abstractEl gen LIS1 es un gen conocido por su importante papel en la migración neuronal durante la etapa del desarrollo embrionario, pero cuando presenta mutaciones da lugar a un trastorno del neurodesarrollo denominado lisencefalia. Se han llevado a cabo amplios estudios en el cerebro, pero este trabajo se centrará especialmente en analizar la función implicada del gen LIS1 en el desarrollo, morfología y crecimiento de las células de Purkinje del cerebelo. Para ello, se silencia el gen LIS1 específicamente en aquellas neuronas que expresan CaMKIIa desde el nacimiento. Para realizar el estudio, se utilizó un modelo de ratón mutante que no presentaba el gen LIS1 debido a un sistema Cre-LoxP en el que los ratones P0 se inyectan con tamoxifeno, lo que activa una enzima llamada Cre que silencia el gen LIS1 únicamente en aquellas neuronas de Purkinje que expresen CaMKIIa y, a través de un estudio morfológico y sináptico mediante microscopía confocal y el programa ImageJ, se espera observar ciertas diferencias significativas a nivel estructural entre los ratones mutantes y wild type, como cambios en la ramificación dendrítica o en la excitación sináptica. Estos cambios y diferencias podrían indicar que el gen LIS1 no solo puede estar implicado en la migración neuronal durante el desarrollo embrionario, sino también en el crecimiento, desarrollo y maduración de las neuronas de Purkinje cerebelosas tras el nacimiento.es_ES
dc.description.abstractThe LIS1 gene is a gene known for its important role in neuronal migration during the embryonic development stage, but when it has mutations, it leads to a neurodevelopmental disorder called lissencephaly. Extensive studies have been carried out in the brain, but this work will focus especially on analyzing the involved function of the LIS1 gene in the development, morphology, and growth of Purkinje cells in the cerebellum. For this, the LIS1 gene will be silenced specifically in those neurons that express CaMKIIa from birth. For this, the LIS1 gene is specifically silenced in those neurons that express CaMKIIa from birth. To carry out the study, a mutant mouse model that lacked the LIS1 gene was used, due to a Cre-LoxP system in which P0 mice are injected with tamoxifen, which activates an enzyme called Cre that silences the LIS1 gene only in those Purkinje neurons that express CaMKIIa. Through a morphological and synaptic study using confocal microscopy and the ImageJ program, certain significant structural differences are expected to be observed between the mutant and wild type mice, such as changes in dendritic branching or in synaptic excitation. These changes and differences could indicate that the LIS1 gene might not only be involved in neuronal migration during embryonic development but also in the growth, development, and maturation of cerebellar Purkinje neurons after birth.es_ES
