dc.description.abstractCiertos compuestos organofosforados (OPs) se emplean como insecticidas y biocidas, dirigidos específicamente contra la enzima acetilcolinesterasa (AChE). En su forma recombinante humana (hrAChE), esta enzima exhibe actividad tanto de fenilacetato esterasa (FAc esterasa) como de fenilvalerato esterasa (PVasa). Asimismo, se ha observado que al incubar la enzima con acetiltiocolina (AtCh), se incrementa su actividad PVasa. La hipótesis actual sugiere un mecanismo tipo “caballo de Troya”, donde la tiocolina liberada en la reacción modifica estructuralmente el centro activo. En trabajos previos observaron diferencias de la sensibilidad en la inhibición con mipafox de la actividad PVasa en muestras pretratadas y sin pretratar. Este trabajo se corrobora las observaciones mostradas en trabajos anteriores y demuestra como la modificación del centro activo por la tCh también puede alterar la inhibición FAc esterasa producida por algunos compuestos organofosforados fosforamidatos. Aportando de esta manera que la naturaleza del inhibidor y el sustrato son muy importantes para explicar las diferencias en muestras pretratadas y no con AtCh.es_ES
dc.description.abstractCertain organophosphorus compounds (OPs) are used as insecticides and biocides, specifically targeting the enzyme acetylcholinesterase (AChE). In its recombinant human form (hrAChE), this enzyme exhibits both phenylacetate esterase (FAc esterase) and phenylvalerate esterase (PVasa) activity. Furthermore, it has been observed that incubating the enzyme with acetylthiocoline (AtCh) increases its PVasa activity. The current hypothesis suggests a ‘Trojan horse’-type mechanism, whereby the thiocoline released during the reaction structurally modifies the active site. Previous studies observed differences in the sensitivity of PVasa activity to inhibition by mipafox between pre-treated and untreated samples. This study corroborates the observations reported in previous work and demonstrates how the modification of the active site by AtCh can also alter the inhibition of FAc esterase produced by certain organophosphorus phosphoramidate compounds. It thus contributes to the understanding that the nature of the inhibitor and the substrate are crucial in explaining the differences between samples pre-treated and not pre-treated with AtCh.es_ES
