Today, antimicrobial resistance (AMR) represents a major global challenge that threatens public health, to the point that it could become the leading cause of death by 2050, surpassing cancer. However, this is not a recent phenomenon. Since the late 1990s, the European Union has already recognised it as an emerging threat. Since then, coordinated strategies have been developed at both international and European level to curb its progression. In this context, Spain established its own National Plan against Antibiotic Resistance (PRAN), following the One Health approach, which recognises that AMR jointly affects humans, animals, and the environment. The aim of this study is to analyse Spain’s response to AMR through the examination of the PRAN 2025–2027 and its contextualisation within the European regulatory framework. To this end, a narrative literature review was conducted based on a progressive search starting from the PRAN itself and the related legislation. Overall, the analysis shows that the PRAN is aligned with European and international strategies under the One Health approach and has become Spain’s main instrument to address AMR. However, the plan still presents certain limitations, such as the lack of its own regulatory framework, heterogeneity in the implementation of measures, uneven development of the One Health approach—particularly in the environmental field—and the need to improve surveillance systems. Therefore, although it has proven to be effective, it needs to continue evolving and strengthening these aspects. In this regard, the potential future approval of a Royal Decree aimed at providing specific legal coverage and regulation for the plan could significantly improve its implementation, coordination, and consistency.es_ES
