The Arabidopsis INCURVATA11 (ICU11) and CUPULIFORMIS2 (CP2) genes are functionally redundant paralogs. It is known that the ICU11 protein—and assumed that CP2 as well—contributes to the transition between the activating epigenetic mark H3K36me3 and the repressive mark H3K27me3. In the laboratory of J.L. Micol, a yeast two-hybrid assay was conducted to identify potential interactors of ICU11, identifying three related to the S-adenosyl-L-methionine (SAM) cycle: ADENOSINE KINASE 2 (ADK2), COBALAMIN-INDEPENDENT METHIONINE SYNTHASE (CIMS), and DEREPRESSED FOR RIBOSOMAL PROTEIN S14 EXPRESSION 2 (DRE2). To circumvent the inconveniences arising from the lethality of null adk1 adk2 double mutants and cims and dre2 single mutants, transgenes producing artificial microRNAs were generated and transferred into Arabidopsis, to partially silence ADK1 and ADK2, CIMS, or DRE2, and to cosilence these genes with ICU11. In this Master's thesis, we characterized the morphological phenotype of these transgenic lines concluding that ICU11 genetically interacts with ADK1 and ADK2. We also compared the published transcriptomic profiles of the atms1-1 and dre2-4 mutants with those of icu11-5 and icu11-5 cp2-1 to identify concordantly deregulated genes and to test the hypothesis of a functional relationship of CIMS and DRE2 with ICU11.
