Ribosome biogenesis is an essential and complex process that occurs in a highly regulated manner in all cells of any living organism. Ribosomal RNA Processing 8 (Rrp8p) is a ribosomal RNA methyltransferase in yeast and humans. With this Master's thesis we have contributed to the characterization of RRP8 in Arabidopsis. We have predicted in silico the function and suborganular localization of RRP8, confirmed the allelism of rrp8-1 and rrp8-2, two insertional alleles that disrupt the RRP8 gene, contributed to the characterization of transgenic lines for RRP8 overexpression and the rescue of its mutant alleles, and initiated the construction of a subcellular localization transgene of the RRP8 protein. We have obtained double mutants between rrp8 mutants and other loss-of-function mutants in genes involved in ribosome biogenesis. We have also analyzed 45S rDNA expression in rrp8 mutants and other mutants affected in genes involved in ribosome biogenesis. On the other hand, we have developed a method based on digital PCR (dPCR) to estimate the copy number of 45S rDNA present in several wild-type accessions and in mutant strains, using the fas1 mutants as control, since they have a reduction in the number of copies of this gene respect to their wild-type backgrounds.
