The pomegranate tree (Punica granatum L.) is widely known as extremely resistant to an abiotic stress such as drought or salinity conditions. However, its behavior under flooding stress and the effect of anaerobic root conditions have not yet been studied. The experiment was carried out with the three most used cultivars in the southeast of Spain: ‘Mollar de Elche’, ‘Valenciana’, and ‘Wonderful’. Pomegranate cuttings were grown in a fertilized substrate under greenhouse conditions. After three weeks, 6 plants of each cultivar had been submerged in pots with fresh water for six days. Biochemical parameters like organic solutes, oxidative stress and hormonal response were determined in leaf and root samples. Flooding increased organic solutes concentration in leaf tissues mainly in ‘Valenciana’ as proline or carbohydrates whilst QAC increase was no significant. However, ‘Valenciana’ hormonal response was the lower in comparison with ‘Mollar de Elche’ and ‘Wonderful’. In addition ‘Valenciana’ was the only one that showed an increase in H2O2 despite any cultivar had significant differences in MDA under flooding conditions. This study agreed with previous physiologic experiment which showed for the first time the low tolerance of these three cultivars due to hypoxic and anoxic conditions
