| dc.description.abstract | El glioblastoma es el tumor primario más frecuente y agresivo del sistema nervioso central, caracterizado por su elevada heterogeneidad y naturaleza infiltrante que limita la eficacia de los tratamientos convencionales. Ante la necesidad de nuevas estrategias terapéuticas, este trabajo evalúa un sistema macromolecular basado en el polímero PMVEMA funcionalizado con la metaloporfirina TPP/NH2-Zn para su aplicación en terapia fotodinámica. La caracterización fotofísica mediante espectroscopia UV-Vis y FTIR confirmó la unión covalente del TPP/NH2-Zn al polímero, manteniendo sus propiedades tras la funcionalización. De forma complementaria, el análisis de fluorescencia mostró modificaciones en las propiedades ópticas, destacando un desplazamiento hipsocrómico en la emisión para el PMVEMA-TPP/NH2-Zn. La evaluación de la viabilidad celular en la línea HGUE-GB-42 mediante ensayos WST-1 y MTT mostró la ausencia de toxicidad en condiciones de oscuridad. Por otra parte, los estudios de internalización celular a 30 min y 2 h demostraron una captación celular dependiente de la dosis y el tiempo, así como la persistencia intracelular del PMVEMATPP/ NH2-Zn tras 24, 48 y 72 h. Además, se confirmó su internalización en modelos 3D de esferoides. En conclusión, el PMVEMA-TPP/NH2-Zn constituye una plataforma biocompatible y prometedora como fotosensibilizador para el tratamiento del glioblastoma. | es_ES |
| dc.description.abstract | Glioblastoma is the most frequent and aggressive primary tumor of the central nervous system, characterized by high heterogeneity and an infiltrative nature that limits the effectiveness of conventional treatments. Given the need for new therapeutic strategies, this work evaluates a macromolecular system based on the polymer PMVEMA functionalized with the metalloporphyrin TPP/NH2-Zn for its application in photodynamic therapy. Photophysical characterization by UV-Vis and FTIR spectroscopy confirmed the covalent binding of TPP/NH2- Zn to the polymer, preserving its properties after functionalization. Complementarily, fluorescence analysis revealed modifications in the optical properties, highlighting a hypsochromic shift in the emission of PMVEMA-TPP/NH2-Zn. Cell viability evaluation in the HGUE-GB-42 cell line using WST-1 and MTT assays showed the absence of cytotoxicity under dark conditions. Furthermore, cellular internalization studies at 30 min and 2 h demonstrated dose and time-dependent uptake, as well as intracellular persistence of PMVEMA-TPP/NH2- Zn after 24, 48, and 72 h. In addition, its internalization was confirmed in 3D spheroid models. In conclusion, PMVEMA-TPP/NH2-Zn constitutes a biocompatible and promising platform as a photosensitizer for glioblastoma treatment. | es_ES |
