Pain is an unpleasant sensory and emotional feeling that alerts the organism against harmful stimuli. Nociceptors are neurons that respond to noxious stimuli and their activation can result in pain. The excitability of nociceptors is regulated by the activity of different ion channels. In the past few years, nociceptors and the ion channels they express have become targets for the development of new analgesic drugs. Due to the difficulties in the experimentation with primary cultures of dorsal root ganglia (DRG), new systems are being developed that allow in vitro studies.
In this work, MED17.11 cells have been differentiated to nociceptors through differentiation protocols. The expression of mRNA and its protein product of neuronal targets like TRPV1, TRKA, CGRP, sodium and potassium channels have been observed through RT-PCR and Western blot techniques. Furthermore, TRPV1 and TrkA were observed in the cells along their differentiation. The functional expression of TRPV1 in MED17.11 cells has been studies through fluorescent calcium images techniques.
The mRNA and protein products of several neuronal targets have been successfully observed. However, calcium imaging studies have shown that the TRPV1 as expressed by the MED17.11 cells was not functional. Furthermore, the cells were not morphologically homogeneous during the different differentiation days. These results suggests that the cells did not differentiate correctly and the differentiation protocol needs to be changed in order to obtain successfully differentiated MED17.11 cells
