Type 2 diabetes mellitus is characterized by the patient's inability to regulate blood glucose levels, which may be due to dysfunction in insulin secretion by pancreatic β-cells or insulin resistance in target tissues. It has been observed that 17-β estradiol (E2) increases glucose-stimulated insulin secretion in pancreatic islets of Langerhans by acting through estrogen receptors. Additionally, estrogens are associated with improved insulin sensitivity in skeletal muscle, liver, and adipose tissue. In this research work, the expression and localization of the estrogen receptor ERβ in the nucleus, Golgi complex, endoplasmic reticulum, and mitochondria in the rat pancreatic β-cell line INS-1E were studied. Furthermore, we have established the silencing of ERβ, via siRNAs, and overexpression of ERβ, through an overexpression plasmid, for potential future studies of loss and gain of function. We have studied how these techniques affect the cellular localization of ERβ in INS-1E cells, obtaining significant findings regarding its subcellular distribution.
