INTRODUCTION: The Central Nervous System (CNS) is the control center of the human body. Understanding its embryonic development is crucial for addressing issues in health, neuroscience, and human behavior. It is important to highlight the significance of neuronal migrations, especially tangential migrations projected from the prethalamic eminence (PThE), and the key neurotransmitters involved in neuronal communication, focusing on glutamate and gamma-aminobutyric acid (GABA). HYPOTHESIS AND OBJECTIVES: Given the existence of tangentially migrated neurons from the PThE, the neurotransmitter nature of migrated neurons in transgenic mice will be investigated, assessing the neurotransmitters contained by Eomes positive neurons. Additionally, the timing of appearance of these cells will be determined. MATERIALS AND METHODS: Three lines of transgenic mice were used to investigate CNS embryonic development. Immunofluorescence was performed on brain sections of EomesCret/flox mice to determine the timing of appearance of Eomes positive neurons during embryonic development with labeling for Eomes (GFPn) along with GABA. Triple immunofluorescence was performed on EomesmTnG mice with labeling for Eomes (GFPn, RFPc) along with GABA or glutamate. RESULTS: Analysis of brain sections from E18.5 EomesmTnG mice reveals the distribution of Eomes positive neurons and the expression of GABAergic and glutamatergic markers in different brain regions, including the cortex, septum, hippocampus, and mesencephalon. Marked presence of Eomes neurons is observed in the cortex and hippocampus, as well as populations of GABAergic neurons in areas such as the striatum and reticular formation. Additionally, expression patterns of Eomes are compared at different stages of embryonic development, highlighting changes in its distribution over time. DISCUSSION: Eomes positive neurons, originating mainly from the PThE, exhibit a diversity of neurotransmitters, tangentially migrating to various brain regions. This phenomenon reflects the heterogeneity observed in other tangential migrations, with both glutamatergic and GABAergic populations. The expression of the Eomes gene in the embryonic development of the mouse CNS progressively decreases with significant variations depending on the brain region and the timing of Cre injection. Late migrations of Eomes positive populations to regions such as the hippocampus and mesencephalic reticular formation are identified. CONCLUSION: Eomes positive neurons, mostly derived from the PThE, display neurotransmitter diversity, reflecting the variability observed in other tangential neuronal migrations. Additionally, the expression of the Eomes gene decreases during embryonic CNS development in mice, with significant variations in its distribution depending on the brain region and the timing of Cre injection.
