Alpha-synuclein (α-syn) is a protein composed of 140 amino acids and encoded by the
SNCA gene, and its main function is the transport and stabilization of synaptic vesicles,
mainly in dopaminergic neurons. α-syn has been found to play an important neurotoxic role
in the pathogenesis of PD, also being the major component of Lewy bodies. α-syn exists in
equilibrium in its soluble forms and in its membrane-bound conformations. However, an
alteration of this equilibrium causes it to aggregate forming oligomers, which are the main
neurotoxic species of α-syn and which, through mechanisms of mitochondrial, metabolic,
and inflammatory response alteration lead to cell injury and death.
The α-syn can spread in a prion-like manner, therefore pathological forms of α-syn can be
detected in many tissues outside the CNS. Different studies seek the best method and
location of measuring α-syn levels in order to have an effective diagnostic method to detect
PD. Naturally, α-syn is also the focus of numerous investigations with the objective of being
able to treat the disease in a curative way by reducing its aggregation, increasing its
degradation or limiting its spread to other cells. Studies focused on diagnostic methods and
those focused on curative therapies are having promising results, but further research is
needed in this regard.
