Liquid biopsy is a potential alternative to tissue biopsy, accounting great advantages: it is a non-invasive process, offers a short response time and allows to determine the tumor molecular profile before and during treatment, during the following-up of the patient and at disease progression. Several studies demonstrate that circulating tumor DNA (ctDNA) is a useful recurrence and survival predictor after surgery and adjuvant chemotherapy in stage II and II colorectal cancer patients.
AIM. The aim of this study is to determine the number of copies of KRAS and BRAF genes mutated allele in ctDNA at diagnosis and after surgery and study its correlation to overall survival (OS), disease-free survival (DFS) and recurrence-free survival (RFS) in patients with stage II and III colorectal cancer.
HYPOTHESIS. ctDNA is a valid prognostic biomarker in patients with stage II and III colorectal cancer, allowing patients stratification according to the risk of recurrence and mortality, so it could be used to guide therapeutical choice during patient management. METHODOLOGY. Observational prospective study of data obtained from the analysis of exon 12 and 13 KRAS and V600F BRAF mutations using digital PCR in doubled-centrifuged plasma samples from stage II and III colorectal cancer patients at diagnosis and after surgery, at University General Hospital Santa Lucia in Cartagena, from January to December 2022.
