Abstract:
Estudios previos con la línea celular EPC de carpa (cipriniformes), persistentemente infectada con IPNV (EPCIPNV), mostraron que dicha línea es resistente a la infección por virus heterólogos, como VHSV, y que secreta al medio factores con actividad antiviral. En el presente trabajo se perseguía obtener una línea celular persistentemente infectada con IPNV de un grupo distinto de peces (perciformes) y para ello se utilizó una línea celular de dorada (SAFIPNV). Se comprobó que sus características son similares a las de EPCIPNV en cuanto a resistencia a superinfección con VHSV, si bien es una línea menos estable. Además se ha constatado la presencia de factores antivirales en los medios condicionados de EPCIPNV y SAFIPNV que inducen resistencia a la infección con VHSV. El análisis por PCR cuantitativa indicó que es probable que la expresión gen mx, inducido por Interferón, pueda tener un papel en el establecimiento del estado antiviral, posiblemente como resultado de la activación de las vías de señalización de Interferón tipo I. Estos estudios permiten profundizar en los mecanismos de la respuesta inmune innata, lo que podría contribuir a la creación y mejora de vacunas frente a virus de peces.
Previous studies with the cell line EPC from carp (cyprinid), persistently infected with IPNV (EPCIPNV), revealed that this line is resistant to heterologous viruses infection, such as VHSV, and that these cells secrete factors with antiviral activity. In this project, the objective was to obtain another cell line persistently infected with IPNV from a different group of fish. To do that, a cell line from gilthead sea bream (perciform) was used (SAFIPNV). It was found that, although this line is less stable, its behavior related to VHSV superinfection resistance is similar to those from EPCIPNV. Moreover, here we show experimental evidence of the presence of antiviral factors in the supernatant of EPCIPNV y SAFIPNV cultures that induce resistance to VHSV infection. Quantitative PCR analysis pointed that mx gene, induced by Interferon, could play a role in the establishment of the antiviral state, possibly through the Interferon type I system. This studies help understand the innate immune response mechanisms, which would contribute to the development and improvement of vaccines against fish viruses’.
